Had this issue when moving an ISO SR from a soon to be decommissioned server to its new location. When trying to attach the SR using the new path, it would give an error "Unable to mount the directory specified in device configuration request" when using XenCenter 5.6.1 (build 38895) on Win7 x64, might be on earlier versions but do not recall it happening before.
Anyways when creating the CIFS, check off 'use different user name' and fill out an appropriate account, even it will be the same one as the current logged in user. For some reason it appears pass through does not work/not implemented.

Check your network location for your CIFS library, not ping your comp, but try to get to destination catalog direc through network and try copy something.
